Acute Toxicity Test for Relazovac™

(Maximum Tolerance Measurement)

Abstract:

Mice were given (ig) Relazovac™ extract three times daily.  The total amount of the compound was 60g/kg (equivalent to 461.54g/kg crude drugs), which was 1282.05 times the clinical dosage (18g/day) for adult.

 Objective:

 D₅₀ can not be measured because of the low toxicity of Relazovac™; however, its toxicity can be reflected through the measurement of a day’s maximum tolerance.

Materials:

1.   Relazovac™, provided by LIFEnhance, Inc., is a brown extract, with batch number 20020402. 0.13g extract is equal to 1g crude drugs.  It is mixed with distilled water into the concentration of 100% and stored in refrigerator at 4°C for use.

2.   The Animals, provided by Hubei Animal Experiment Center , were Kunming-breed mice with the weight of 19±1g, and with the same number of ♂ (male) ♀ (female).  The mouse food was purchased from the same center.

Methods:

According to the requirement of the literature [1], 22 Kunming-breed mice, with the weight of 19±1g, and the same number of ♂♀, were given nothing but water for 16 hours.  They were given 20g of the extract at a time (20ml/kg) three times daily and then were observed at least once a day for 7 consecutive days to see their reaction.

Results: 

No mice died after taking the compound and no abnormal phenomena was observed in their activities except the following: their excrement softened and darkened in color.  They ate more after being given the extract two times. But the next day, all the activities became normal.  According to the formula in the literature [2], the tolerance times is calculated as follows.

                                          Per mice’s tolerance     adult average weight (50,000g)

The mice’s tolerance times=—————————×—————————————

                                          Average weight (20g)      adult’s day’s dosage

                                               9.23g×3             50000g

=————         ×     ————      =1282.05 times

                                                   20g               18g

20g/Kg ×3=60g/kg medicine, equal to 461.54g/kg crude drugs.  The mice tolerance is 1282.05 times the adult clinical dosage.
